Yes, it's possible. Use the mergesis command like this:
Code:
ensymble.py mergesis sisfile1.sis sisfile2.sis ... sisfilen.sis resultingfile.sis
This inserts all the files that follow the first one into the first one, and its certificate is not preserved. Read Ensymble's reamde file, the mergesis section, for more information.

I want to put inside of program.sis main the .sis of appuifw2 and uitricks; I would have to make thus?

You would have to specify the name of the resulting file too.

ensymble.py mergesis program.sis appuifw2.sis uitricks.sis program_final.sis
program_final.sis now contains program.sis, appuifw2.sis and uitricks.sis.

Therefore program_final.sis it corresponds to the .sis of the program that I will go to install on the cellular one?

But ensymble is compatible also for the 2nd edition?

Ensymble-generated packages won't work on 2nd edition phones. Although, you can use py2sisng instead
